动物克隆的分子机制及衰老相关基因的克隆

 动物克隆分子机制研究
克隆技术在农业育种和生物医学等领域具有广泛的应用前景,但是迄今为止,无论是胚胎细胞克隆还是体细胞克隆,克隆效率都较低(1%-3%),即使少量出生的个体也常常伴有一些系统或器官的异常发育。因此探索克隆效率低下的原因,研究克隆的分子作用机理已成为当前该领域研究的热点。一般认为,体细胞核移入去核的卵细胞后要发生再程序化(Reprogramming)过程,而再程序受阻或再程序不完全可能导致重构胚发育的各种后果;因此研究植入前克隆胚胎的基因表达模式对提高克隆效率和减少畸形的发生有重要意义。
体细胞克隆动物分子机制的研究已取得重要进展,成功地建立了DD-RT-PCR(差异显示逆转录PCR)和单细胞减法PCR构建cDNA文库(Subtracted PCR-Generated cDNA Libraries)技术系统; 现已获得80多条兔早期克隆胚再程序化相关差显片段和30多条与小鼠早期胚发育相关差显片段,完成了对部分差显片段的鉴定和分析工作; 克隆了3条全长cDNA基因, 其中包括次从兔卵巢中克隆到了全长的LAPTM4αcDNA序列,并利用原位杂交技术就其在卵泡发育成熟过程中的定位表达进行了系统的研究。在卵泡发育成熟过程中,LAPTM4α在晚期生长卵泡、成熟卵泡的颗粒细胞以及晚期生长卵泡靠近卵巢表面的组织中具有特异地表达的特性(Fig.1),这表明:LAPTM4α可能与卵泡的成熟以及排卵过程直接相关,在胚胎基因组激活时的特异表达说明它在母源转化以及胚胎自主发育的过程中也起着重要的作用。

二, 衰老及中药抗衰老研究
     本课题使用DDRT-PCR方法研究衰老鼠小鼠(SAM)脑组织的基因表达情况,比较不同年龄间的小鼠脑组织基因的表达情况及同龄不同品系小鼠间脑组织的基因表达情况; 并对使用中医药物对基因表达的影响情况进行了比较。己分离获得了95个阳性差异表达片段。经初步分析鉴定,确认衰老相关的基因片段, 筛选其全长的基因序列; 当前本课题组己克隆了两个带有开放读框的cDNA片段,其中一个是含有完整的阅读框。这为下一步开展衰老相关基因功能的研究打下了基础。
